SI-109 is a novel and potent inhibitor of STAT3 SH2 domain with potential antitumor activity. It inhibits STAT3 with a Ki of 9 nM. SI-109 exhibited antitumor activity by effectively inhibiting the transcriptional activity of STAT3 with an IC50 of3 μM. SI-109 along with the CRBN ligand lenalidomide can be used to construct PROTAC STAT3 degrader SD-36.
