背景信息：

DNA 结合/分化 (ID) 蛋白的抑制剂是旨在抑制碱性螺旋-环-螺旋 (bHLH) 转录因子活性的一个蛋白家族。在人体中发现了 4 种 ID 蛋白 (ID1-4)，它们都含有一个螺旋-环-螺旋结构域，但缺乏碱性 DNA 结合结构域。用 bHLH 转录因子结合形成的异源二聚体可以减弱 bHLH 蛋白活性并阻止它们与 DNA 结合 (1)。ID 蛋白在发育过程中发挥着重要作用，主要抑制干细胞/祖细胞提前成熟分化 (1,2)。ID3 在免疫系统发育过程中发挥着重要的作用，可以抑制 E2A 介导的 T 细胞分化 (3)。针对小鼠模型的研究已经证明，ID3 同合型缺失会阻断调节性 T 细胞分化(4)并导致 γδ T 细胞淋巴瘤的产生 (5)。已证明在造血室外部 ID3 能抑制 MyoD，表明 ID3 与 TGFβ 介导的肌肉修复有关 (6)。同样，研究表明 ID3 能抑制结肠癌细胞中的 p21，据称此功能能够增强可能的癌起始细胞的自我更新能力 (7)。
