产品介绍This gene is a member of the glycoprotein hormone beta chain family and encodes the beta 3 subunit of chorionic gonadotropin (CG). Glycoprotein hormones are heterodimers consisting of a common alpha subunit and an unique beta subunit which confers biological specificity. CG is produced by the trophoblastic cells of the placenta and stimulates the ovaries to synthesize the steroids that are essential for the maintenance of pregnancy. The beta subunit of CG is encoded by 6 genes which are arranged in tandem and inverted pairs on chromosome 19q13.3 and contiguous with the luteinizing hormone beta subunit gene. 靶点与功能

hCG是由胎盘滋养层细胞分泌的糖蛋白激素，由α亚基（92个氨基酸，与LH、FSH、TSH共享）和β亚基（145个氨基酸，含28-30个唾液酸残基） 组成，β亚基的羧基端26个氨基酸为hCG特有，是特异性识别的关键区域。其核心功能包括：

生理作用：维持黄体功能（促进孕酮分泌），支持早期妊娠；

临床价值：血清hCGβ水平是早孕诊断、宫外孕排查及妊娠滋养细胞疾病（如葡萄胎、绒癌）的金标准标志物，正常妊娠早期每48小时翻倍，异常妊娠时增长缓慢或下降；

研究意义：在肿瘤领域，hCGβ是非妊娠女性生殖细胞肿瘤（如卵巢无性细胞瘤）和某些腺癌（如肺癌、膀胱癌）的异位标志物。

实验操作关键要点

ELISA定量检测（血清/尿液样本）：

双抗夹心体系：推荐兔单抗作为检测抗体（1μg/mL），小鼠单抗作为捕获抗体（2μg/mL），可避免交叉反应；

样本处理：血清需1:100稀释（含1% BSA的PBS），尿液样本需经0.22μm滤膜过滤（去除结晶干扰）；

灵敏度：优质抗体可检测低至2mIU/mL的hCGβ（满足早孕诊断需求，临床阈值为>5mIU/mL）。

IHC-P组织染色（胎盘或肿瘤样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复10分钟（针对福尔马林固定的石蜡切片），增强β亚基抗原暴露；

阳性定位：胎盘绒毛滋养层细胞胞质呈棕黄色颗粒状阳性，绒癌细胞中弥漫强阳性；

阴性对照：非妊娠子宫内膜组织（无信号）或添加hCGβ重组蛋白封闭抗体（信号消失）。

WB验证（JEG-3细胞裂解液）：

样本制备：使用含蛋白酶抑制剂（1mM PMSF）和糖酶（如PNGase F，37℃孵育1小时） 的RIPA裂解液，去除糖基化干扰；

电泳条件：15% SDS-PAGE凝胶（分离小分子蛋白），转膜采用0.22μm PVDF膜（200mA恒流45分钟）；

抗体孵育：兔单抗1:2000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:10000室温孵育1小时。
