产品介绍The four human glycoprotein hormones chorionic gonadotropin (CG), luteinizing hormone (LH), follicle stimulating hormone (FSH), and thyroid stimulating hormone (TSH) are dimers consisting of alpha and beta subunits that are associated noncovalently. The alpha subunits of these hormones are identical, however, their beta chains are unique and confer biological specificity. The protein encoded by this gene is the alpha subunit and belongs to the glycoprotein hormones alpha chain family. Two transcript variants encoding different isoforms have been found for this gene. 靶点与功能

HCGα是由145个氨基酸组成的糖蛋白亚基，分子量约14kDa，与垂体分泌的FSH、LH、TSH的α亚基高度同源（约80%序列一致），因此易产生交叉反应。其核心功能包括：

临床价值：作为早孕试纸、ELISA检测试剂盒的核心试剂，通过识别HCGα亚基实现妊娠诊断；

研究意义：辅助鉴别滋养细胞肿瘤（如葡萄胎、绒毛膜癌），因这类肿瘤会异常分泌HCGα游离亚基；

交叉反应特性：需通过抗体设计避免与FSH/LH/TSH的α亚基交叉，确保检测特异性。

实验操作关键要点

ELISA双抗体夹心法（早孕检测）：

包被抗体：选择高亲和力HCGα单抗（如4A8），用0.05M碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜；

检测抗体：生物素标记的HCGβ单抗（避免α亚基交叉），1:1000稀释后37℃孵育1小时，结合HRP-链霉亲和素显色；

特异性控制：需设置FSH/LH/TSH阳性对照孔，确保无交叉反应（OD值＜0.1）。

IHC-P组织染色（滋养细胞肿瘤样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟，冷却至室温；

阳性定位：肿瘤细胞胞质/胞膜呈棕黄色颗粒状阳性，正常胎盘组织对照呈弱阳性；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭30分钟，减少非特异性结合。
