产品介绍The protein encoded by this gene is a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is found in secretory vesicles of neurons and endocrine cells. This gene product is a precursor to three biologically active peptides; vasostatin, pancreastatin, and parastatin. These peptides act as autocrine or paracrine negative modulators of the neuroendocrine system. Other peptides, including chromostatin, beta-granin, WE-14 and GE-25, are also derived from the full-length protein. However, biological activities for these molecules have not been shown. 靶点与功能

嗜铬粒素A（Chromogranin A, CgA）是一种酸性可溶性糖蛋白，分子量约49kDa，主要存在于神经内分泌细胞的分泌颗粒中。其核心功能包括：

生理作用：参与神经内分泌颗粒的组装与分泌调节；

临床价值：作为神经内分泌肿瘤（NETs）的**标志物，血清CgA水平与肿瘤负荷、转移风险正相关，正常参考值≤100ng/mL；

诊断意义：通过IHC检测肿瘤组织中CgA的表达，可辅助区分NETs与非神经内分泌肿瘤（如肺癌、胃肠道癌）。

实验操作关键要点

IHC-P病理诊断（石蜡切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），避免高温破坏CgA抗原表位；

阳性定位：神经内分泌细胞胞质呈棕黄色颗粒状阳性，嗜铬细胞瘤中呈弥漫强阳性，肺类癌中呈灶状分布；

对照设置：必须包含正常肾上腺髓质组织（阳性对照）和肝细胞组织（阴性对照）。

WB验证（肿瘤细胞裂解液）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：一抗1:1500稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠/兔）1:5000室温孵育1小时。
