产品介绍The serine-threonine protein kinase encoded by the AKT1 gene is catalytically inactive in serum-starved primary and immortalized fibroblasts. AKT1 and the related AKT2 are activated by platelet-derived growth factor. The activation is rapid and specific, and it is abrogated by mutations in the pleckstrin homology domain of AKT1. It was shown that the activation occurs through phosphatidylinositol 3-kinase. In the developing nervous system AKT is a critical mediator of growth factor-induced neuronal survival. Survival factors can suppress apoptosis in a transcription-independent manner by activating the serine/threonine kinase AKT1, which then phosphorylates and inactivates components of the apoptotic machinery. Mutations in this gene have been associated with the Proteus syndrome. Multiple alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Jul 2011]

Akt（蛋白激酶B）是丝氨酸/苏氨酸激酶家族成员（包括Akt1/Akt2/Akt3三个亚型），核心功能与研究价值包括：

信号通路枢纽：通过磷酸化（Ser473/Thr308位点）激活下游靶蛋白（如mTOR、GSK3β），调控细胞存活（抑制凋亡）、糖代谢（促进GLUT4转位）及肿瘤侵袭（上调MMPs表达）；

疾病标志物：在乳腺癌、前列腺癌等实体瘤中Akt持续激活（p-Akt阳性率>60%），与化疗耐药密切相关，是靶向药物开发的关键靶点；

实验操作关键要点

WB通路活性检测：

样本处理：HepG2细胞用含磷酸酶抑制剂（PhosSTOP） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

抗体配对：总Akt抗体（1:2000）与p-Akt Ser473抗体（1:1000）同步检测，β-actin内参校正，计算p-Akt/Akt比值（LPS刺激后比值升高3-5倍）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳90分钟（确保60kDa条带分离）。

IHC-P肿瘤组织染色：

抗原修复：EDTA缓冲液（pH9.0）高压修复10分钟（石蜡切片），阳性信号定位于癌细胞胞质/胞膜（乳腺癌组织p-Akt阳性率与预后负相关）；

抗原特异性验证：使用Akt抑制剂处理的细胞/组织作为阴性对照（p-Akt信号消失）；

染色优化：抗体稀释至1:300，37℃孵育30分钟，DAB显色3-5分钟（避免过染掩盖弱阳性信号）。

IP互作蛋白筛选：

推荐使用Proteintech Akt1抗体（2μg/样本），4℃旋转孵育过夜，Protein A/G磁珠捕获免疫复合物，洗脱后通过质谱鉴定Akt结合蛋白（如14-3-3蛋白）。
