产品介绍The product encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Mutations in this gene cause nemaline myopathy type 3, congenital myopathy with excess of thin myofilaments, congenital myopathy with cores, and congenital myopathy with fiber-type disproportion, diseases that lead to muscle fiber defects. 肌动蛋白α（ACTA2基因编码）是肌动蛋白家族的平滑肌特异性亚型（分子量约42kDa），核心功能与研究价值包括：

细胞“身份标签”：特异性表达于血管平滑肌、胃肠道平滑肌及活化的肌成纤维细胞，静息态呈“丝状分布”，活化后（如纤维化）形成“应力纤维”，是区分平滑肌与上皮/间质细胞的关键标志物；

病理诊断核心：在平滑肌肿瘤（平滑肌瘤/肉瘤）、肌上皮瘤中高表达，是病理科鉴别肿瘤来源的**抗体（阳性率＞95%）；

纤维化研究枢纽：肝纤维化时肝星状细胞活化并表达α-SMA，与胶原沉积程度正相关，是评估纤维化进展的量化指标（如肝纤维化分期中α-SMA⁺细胞占比每增加10%，纤维化程度升高1期）。

实验操作关键要点

IHC-P病理诊断优化：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），平滑肌细胞胞质呈棕黄色阳性（肿瘤细胞呈弥漫性强阳性，正常平滑肌呈灶性阳性）；

对照设置：必须包含正常子宫平滑肌（阳性对照）和结肠上皮细胞（阴性对照，避免交叉反应）；

染色判读：阳性细胞比例＞10%判定为“平滑肌来源肿瘤”，＜5%为“非平滑肌来源”。

WB纤维化模型验证：

样本处理：CCl₄诱导肝纤维化小鼠肝组织，用含1mM PMSF的RIPA裂解液冰上裂解30分钟；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜至0.45μm PVDF膜；

结果判读：抗体1:2000稀释，4℃孵育过夜，42kDa处显带，纤维化组灰度值为对照组的4-6倍。

IF肌成纤维细胞活化监测：

与波形蛋白（Vimentin） 共染，α-SMA（红色）与Vimentin（绿色）在肌成纤维细胞中“共定位”，形成“纺锤形双阳性细胞”（纤维化组织中占比＞30%提示进展期病变）。
