产品介绍All eukaryotic cells express Actin, which often constitutes as much as 50% of total cellular protein. Actin filaments can form both stable and labile structures and are crucial components of microvilli and the contractile apparatus of muscle cells. While lower eukaryotes, such as yeast, have only one Actin gene, higher eukaryotes have several isoforms encoded by a family of genes. At least six types of Actin are present in mammalian tissues and fall into three classes. alpha-Actin expression is limited to various types of muscle, whereas beta- and gamma-Actin are the principle constituents of filaments in other tissues. Members of the small GTPase family regulate the organization of the Actin cytoskeleton. Rho controls the assembly of Actin stress fibers and focal adhesion. Rac regulates Actin filament accumulation at the plasma membrane. Cdc42 stimulates formation of filopodia.

α-SMA(α-Smooth Muscle Actin)是平滑肌细胞和肌成纤维细胞的主要结构蛋白(分子量42kDa),核心功能与研究价值包括:
纤维化“驱动因子”:在肝星状细胞、肺成纤维细胞活化后高表达,通过促进细胞收缩和细胞外基质(ECM)沉积,推动肝纤维化、肾间质纤维化等疾病进展(α-SMA⁺细胞数量与纤维化程度正相关);
肿瘤微环境标志物:肿瘤相关成纤维细胞(CAF)高表达α-SMA,通过分泌TGF-β、VEGF等因子促进肿瘤血管生成和侵袭转移(乳腺癌CAF中α-SMA表达量是正常成纤维细胞的3-5倍);
血管结构研究:标记血管平滑肌细胞,用于血管新生、动脉粥样硬化等血管疾病模型的形态学分析。
实验操作关键要点
IHC-P肝纤维化检测:
抗原修复:柠檬酸盐缓冲液(pH6.0)微波修复20分钟(石蜡切片),肝纤维化组织中汇管区肌成纤维细胞胞质呈棕黄色阳性(正常肝组织阴性);
阳性对照:大鼠子宫平滑肌组织(平滑肌细胞强阳性);
染色优化:抗体稀释至1:800,37℃孵育30分钟,DAB显色3分钟(避免过染掩盖细胞形态)。
WB肌成纤维细胞活化验证:
样本处理:TGF-β1(10ng/mL)诱导L929成纤维细胞48小时,RIPA裂解液(含1mM PMSF)冰上裂解30分钟;
电泳条件:12% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,电泳90分钟;
结果判读:抗体1:2000稀释,4℃孵育过夜,42kDa处显带,活化组灰度值为对照组的4-6倍。
IF肿瘤微环境共定位:
与FSP-1抗体共染(CAF双标志物),α-SMA(绿色)与FSP-1(红色)在肿瘤间质中双阳性表达,共定位系数>0.6提示CAF高活化状态(乳腺癌组织中常见)。
