靶点与功能

嗜铬粒素B（Chromogranin B, CgB）是一种酪氨酸硫酸化分泌蛋白，由657个氨基酸组成（含20个氨基酸信号肽），分子量约72-76kDa。其核心功能包括：

生理作用：参与神经内分泌细胞分泌颗粒的组装与激素释放调节，可作为其他生物活性肽的前体；

临床价值：作为神经内分泌肿瘤（NENs）的辅助标志物，尤其在CgA表达阴性或低表达时，CgB检测能提高诊断灵敏度；

组织分布：主要在肾上腺、垂体、胸腺、子宫、卵巢等神经内分泌组织高表达。

实验操作关键要点

IHC-P组织染色（神经内分泌肿瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），冷却至室温后用3% H₂O₂封闭内源性过氧化物酶10分钟；

阳性定位：肿瘤细胞胞质呈棕黄色颗粒状阳性，正常神经内分泌组织（如肾上腺髓质）可作为阳性对照；

抗体稀释：推荐1:200稀释（5% BSA+TBST），4℃孵育过夜，HRP标记二抗室温孵育1小时。

WB验证（细胞/组织裂解液）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（300mA恒流2小时）；

结果判断：在72-76kDa处出现特异性条带，阴性对照（如肝细胞裂解液）无条带。
