产品介绍Cytokeratins comprise a diverse group of intermediate filament proteins (IFPs) that are expressed as pairs in both keratinized and non-keratinized epithelial tissue. Cytokeratins play a critical role in differentiation and tissue specialization and function to maintain the overall structural integrity of epithelial cells. Cytokeratins have been found to be useful markers of tissue differentiation which is directly applicable to the characterization of malignant tumors. Cytokeratins 10 and 13 are present in the cytoskeletal region of a subset of squamous cell carcinomas. Cytokeratin 10 is a heterotetramer of two type I and two type II keratins, is generally associated with keratin 1, and is seen in all suprabasal cell layers including stratum corneum.

靶点与功能

细胞角蛋白10（CK10）是I型酸性细胞角蛋白家族成员，分子量约56-59kDa，主要表达于终末分化的角质形成细胞（如皮肤颗粒层、角质层），与CK1形成异二聚体构成细胞中间纤维网络。其核心功能包括：

生理作用：维持上皮细胞机械强度和皮肤屏障完整性，是角质细胞终末分化的关键标志物；

临床价值：作为鳞状细胞癌（如皮肤鳞癌、食管癌） 的诊断标志物，在肿瘤分化程度评估中具有特异性；

疾病关联：基因突变可导致表皮松解性角化过度症（EHK），表现为皮肤水疱和角化异常。

实验操作关键要点

WB检测（皮肤组织/细胞裂解液）：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体稀释：一抗1:2000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光时间30-60秒。

IHC-P组织染色（皮肤鳞癌样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），自然冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞质呈棕黄色颗粒状阳性，正常皮肤颗粒层可作为阳性对照；

封闭优化：5%驴血清+0.3% Triton X-100室温封闭30分钟，减少非特异性结合。
