产品介绍KRT18 encodes the type I intermediate filament chain keratin 18. Keratin 18, together with its filament partner keratin 8, are perhaps the most commonly found members of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

靶点与功能

细胞角蛋白18（Cytokeratin 18, CK18）是II型酸性角蛋白，分子量约45kDa，主要表达于单层上皮细胞（如肝、胰、肺、乳腺导管上皮），与CK8形成异二聚体。其核心功能包括：

生理作用：维持上皮细胞机械稳定性，参与细胞凋亡信号传导（如被caspase切割后释放促凋亡片段）；

临床价值：作为腺癌的特异性标志物（如肝细胞癌、胰腺癌、肺腺癌），与鳞癌标志物（CK5/6、CK17）联合检测可提高肿瘤分型准确率；

研究意义：血清CK18片段（如M30、M65）可作为肝损伤、肿瘤细胞凋亡的非侵入性检测指标。

实验操作关键要点

IHC-P区分腺癌与鳞癌：

抗原修复：EDTA缓冲液（pH9.0） 高压修复3分钟（优于柠檬酸钠缓冲液，适合福尔马林固定样本）；

阳性定位：腺癌组织胞质弥漫性棕黄色阳性，鳞癌组织阴性；肝组织中正常肝细胞呈弱阳性，肝癌细胞强阳性；

对照设置：必须包含肺腺癌（阳性） 和肺鳞癌（阴性） 组织对照。

WB检测凋亡片段（M30）：

样本处理：使用含10μM MG132（蛋白酶体抑制剂）的RIPA裂解液，避免片段降解；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、150V分离胶，转膜用0.22μm PVDF膜；

抗体选择：若检测凋亡片段需使用识别切割位点的单抗（如M30抗体），总CK18检测用全长抗体。
