产品介绍KRT18 encodes the type I intermediate filament chain keratin 18. Keratin 18, together with its filament partner keratin 8, are perhaps the most commonly found members of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

靶点与功能

细胞角蛋白18（CK18）是I型酸性细胞角蛋白，分子量约45kDa，主要表达于单层上皮细胞（如肝细胞、肾小管上皮、胃肠道黏膜上皮），与CK8形成异二聚体构成细胞骨架。其核心功能包括：

生理作用：维持单层上皮细胞形态与机械稳定性，参与细胞凋亡信号传导（如被caspase切割产生凋亡标志物CK18片段）；

临床价值：作为腺癌诊断标志物（如肝癌、胰腺癌、乳腺癌），可通过检测血清CK18片段（M30/M65）评估肿瘤细胞凋亡与坏死程度；

疾病关联：在肝纤维化、非酒精性脂肪肝中表达上调，是肝损伤的敏感指标。

实验操作关键要点

WB检测（肝癌细胞系/组织）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 和磷酸酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（200mA恒流1.5小时）；

抗体稀释：一抗1:5000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:10000室温孵育1小时，曝光时间15-30秒（避免信号过曝）。

IHC-P组织染色（胰腺癌样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟（95℃），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞质呈弥漫性棕黄色阳性，正常胰腺导管上皮可作为阳性对照；

信号增强：对低表达样本，可使用Polymer HRP检测系统（如Dako EnVision+），较传统ABC法信号提升2倍。
