产品介绍Cytokeratin 20 is a type I keratin which is primarily expressed in gastric and intestinal epithelium, urothelium, and Merkel-cells. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Their genes are clustered in a region of chromosome 17q12-q21. Cytokeratin 20 is a major cellular protein of mature enterocytes and goblet cells and is specifically expressed in the gastric and intestinal mucosa. It is also expressed in adenocarcinomas of the colon, stomach, pancreas and the bile system and is present in mucinous ovarian tumors, transitional-cell and Merkel-cell carcinomas. Notably, the squamous cell carcinomas and adenocarcinomas of the breast, lung, and endometrium, non-mucinous tumors of the ovary, and small cell carcinomas lack cytokeratin 20.

靶点与功能

细胞角蛋白20（Cytokeratin 20, CK20）是I型酸性角蛋白，分子量约46kDa，主要表达于胃肠道上皮、尿路上皮及 Merkel 细胞。其核心功能与临床价值包括：

组织特异性：正常情况下仅在小肠、结肠、胃黏膜上皮表达，是胃肠道腺癌的特异性标志物；

肿瘤诊断：结直肠癌（阳性率90%）、胃癌（60-70%）、 Merkel 细胞癌（100%）中高表达，与CK7联合检测可区分原发灶（如CK7⁻/CK20⁺提示结直肠来源）；

预后意义：结直肠癌中CK20表达强度与淋巴结转移呈正相关，可作为预后评估辅助指标。

实验操作关键要点

IHC-P病理诊断（结直肠癌样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复5分钟（针对福尔马林固定石蜡切片），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞质呈棕黄色颗粒状阳性，典型特征为“灶状或散在分布”（与CK7弥漫表达不同）；

抗体稀释：推荐1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP聚合物二抗室温孵育30分钟。

WB验证（细胞裂解液）：

样本选择：阳性对照用结直肠癌细胞系（如HCT116），阴性对照用肺癌细胞系（如A549）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1.5小时）；

结果判断：46kDa处出现单一特异性条带，阴性对照无条带。
