产品介绍The protein encoded by this gene is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. This type II cytokeratin is specifically expressed in the simple epithelia lining the cavities of the internal organs and in the gland ducts and blood vessels. The genes encoding the type II cytokeratins are clustered in a region of chromosome 12q12-q13. Alternative splicing may result in several transcript variants; however, not all variants have been fully described. [provided by RefSeq, Jul 2008]

靶点与功能

CK7是酸性中间丝蛋白家族成员，分子量约40-55kDa，主要表达于乳腺、肺、泌尿系统等单层上皮细胞，在肺癌、乳腺癌、卵巢癌等上皮源性肿瘤中特异性高表达。其功能不仅限于维持细胞结构，还参与细胞信号转导和肿瘤侵袭，是临床病理中鉴别肿瘤原发部位的重要标志物（如CK7+/CK20-常提示肺或卵巢来源肿瘤）。

实验操作关键要点

IHC-P优化（肿瘤组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性对照：推荐使用肺腺癌组织（癌细胞胞质呈棕黄色颗粒状阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。

WB注意事项：

样本处理：RIPA裂解液（含蛋白酶抑制剂）冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约54kDa；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时。
