产品介绍c-Kit is a transmembrane tyrosine kinase encoded by the cKit proto oncogene. c-Kit acts to regulate a variety of biological responses including cell proliferation, apoptosis, chemotaxis and adhesion. Ligand binding to the extracellular domain leads to autophosphorylation on several tyrosine residues within the cytoplasmic domain, and activation. Mutations in c-Kit have been found to be important for tumor growth and progression in a variety of cancers including mast cell diseases, gastrointestinal stromal tumor, acute myeloid leukemia, Ewing sarcoma and lung cancer. Phosphorylation at tyrosine 721 of c-Kit allows binding and activation of PI3 kinase.

靶点与功能

c-kit（CD117）是III型跨膜受体酪氨酸激酶，由KIT基因编码，分子量约145kDa，配体为干细胞因子（SCF）。其核心功能包括：

生理作用：调控造血干细胞分化、肥大细胞发育及胃肠道间质细胞增殖；

临床价值：95%的胃肠道间质瘤（GIST）表达CD117，是诊断GIST的“金标准”，也是靶向药物（如伊马替尼）疗效预测的关键指标；

研究意义：在急性髓系白血病、黑色素瘤等肿瘤中常发生突变（如Exon 11缺失），是肿瘤发生机制研究的重要靶点。

实验操作关键要点

IHC-P优化（GIST病理诊断）：

抗原修复：采用EDTA缓冲液（pH8.0） 高压修复5分钟（针对福尔马林固定石蜡切片），确保胞膜信号清晰；

阳性定位：肿瘤细胞细胞膜/胞质呈棕黄色阳性，正常胃肠道间质细胞弱阳性；

对照设置：必须包含GIST阳性组织（强阳性对照）和平滑肌瘤组织（阴性对照）。

WB检测注意事项：

样本处理：RIPA裂解液（含1mM PMSF和磷酸酶抑制剂）冰上裂解30分钟，12000g离心取上清；

电泳条件：8% SDS-PAGE凝胶（因蛋白分子量较大），恒压80V浓缩胶、120V分离胶，目标条带约145kDa；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
