产品介绍c-Kit is a transmembrane tyrosine kinase encoded by the cKit proto oncogene. c-Kit acts to regulate a variety of biological responses including cell proliferation, apoptosis, chemotaxis and adhesion. Ligand binding to the extracellular domain leads to autophosphorylation on several tyrosine residues within the cytoplasmic domain, and activation. Mutations in c-Kit have been found to be important for tumor growth and progression in a variety of cancers including mast cell diseases, gastrointestinal stromal tumor, acute myeloid leukemia, Ewing sarcoma and lung cancer. Phosphorylation at tyrosine 721 of c-Kit allows binding and activation of PI3 kinase.

靶点与功能

c-kit（CD117）是III型受体酪氨酸激酶，分子量约145kDa，由原癌基因c-kit编码，其配体为干细胞因子（SCF）。核心功能与临床价值包括：

生理作用：调控造血干细胞增殖、肥大细胞分化及黑色素细胞迁移，是胚胎发育和组织稳态的关键分子；

肿瘤关联：在胃肠道间质瘤（GIST，突变率80-85%）、急性髓系白血病（AML）、黑色素瘤中高频突变或过表达，是靶向药物（如伊马替尼）的重要作用靶点；

诊断价值：CD117阳性是GIST的诊断金标准（IHC阳性率>95%），可与平滑肌瘤、神经鞘瘤鉴别。

实验操作关键要点

IHC-P诊断GIST样本：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复5分钟（优于柠檬酸钠缓冲液），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞膜/胞质呈棕黄色颗粒状阳性，典型特征为“膜着色为主，质着色为辅”；

抗体稀释：推荐1:500稀释（5% BSA+TBST），4℃孵育过夜，HRP聚合物二抗室温孵育30分钟，DAB显色3-5分钟。

流式细胞术分选造血干细胞：

样本处理：4% PFA固定15分钟，90%甲醇通透20分钟（破膜后检测胞内/膜蛋白）；

抗体用量：荧光标记抗体1:250稀释（0.1μg/1x10⁶细胞），4℃避光孵育30分钟；

对照设置：需包含同型对照（兔IgG）和未染色对照，排除非特异性结合。
