产品介绍CDC-like kinase 2 (CLK2) belongs to a family of autophosphorylating kinases termed CLK (CDC2/CDC28-like kinases), which have been shown to phosphorylate serine- and arginine-rich (SR) proteins of the spliceosomal complex, and to influence alternative splicing in overexpression systems. Recent findings demonstrated that the CLK kinases activate PTP-1B family members, and this phosphatase may be an important cellular target for CLK action. Mutations in the CLK2 proteins affect organismal features such as development, behavior, reproduction, and aging as well as cellular features such as the cell cycle, apoptosis, the DNA replication checkpoint, and telomere length.

靶点与功能

细胞分裂周期样激酶2（CLK2）是双特异性丝氨酸/苏氨酸-酪氨酸激酶，分子量约50kDa，属于CLK家族成员（包括CLK1-4）。其核心功能包括：

分子机制：磷酸化剪接体复合体中的SR蛋白（如SRSF1、SRSF3），调控前体mRNA选择性剪接；

细胞功能：参与细胞周期检查点激活（如DNA损伤后磷酸化p53，诱导G2/M期阻滞）、端粒长度维持及能量代谢调控（抑制PGC1α转录活性，减少肝糖输出）；

疾病关联：在肺癌、乳腺癌中高表达，与肿瘤细胞耐药性相关；CLK2突变可导致发育异常（如胚胎致死、繁殖缺陷）。

实验操作关键要点

WB检测（肺癌细胞系）：

样本处理：使用含1mM PMSF+磷酸酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体稀释：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒。

IHC-P组织染色（乳腺癌样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞核/胞质共定位（核内与剪接体结合，胞质参与信号传导）；

对照设置：需包含CLK2敲除细胞系（如HEK293T-CLK2-KO）作为阴性对照。
