产品介绍This gene encodes the major type IV alpha collagen chain of basement membranes. Like the other members of the type IV collagen gene family, this gene is organized in a head-to-head conformation with another type IV collagen gene so that each gene pair shares a common promoter. 靶点与功能

IV型胶原蛋白（Collagen IV）是基底膜的主要结构蛋白，由α1-α6六个亚基组成（如α1α1α2、α3α4α5等），分子量约400kDa。核心功能与临床价值包括：

生理作用：构成基底膜骨架，维持组织完整性，参与细胞黏附、迁移及信号传导；

疾病关联：Alport综合征（COL4A3/4/5基因突变）、糖尿病肾病（基底膜增厚）、肿瘤侵袭转移（IV型胶原酶降解）的关键标志物；

研究意义：通过IHC定位基底膜分布，辅助肿瘤血管生成、肾小球损伤等病理机制研究。

实验操作关键要点

IHC-P检测肾小球基底膜：

抗原修复：采用胃蛋白酶消化法（0.4%胃蛋白酶，37℃孵育30分钟），优于常规柠檬酸钠修复（IV型胶原交联紧密）；

阳性定位：肾小球基底膜、肾小管基底膜及血管基底膜呈连续线性棕黄色阳性，Alport综合征样本可见断裂或缺失；

抗体稀释：推荐1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP聚合物二抗室温孵育30分钟。

WB验证（基底膜提取物）：

样本处理：使用含1M urea的RIPA裂解液（破坏胶原三螺旋结构），冰上裂解60分钟，12000g离心20分钟取上清；

电泳条件：7.5% SDS-PAGE凝胶（低浓度胶分离大分子量蛋白），恒压60V浓缩胶、80V分离胶，转膜用0.45μm硝酸纤维素膜；

结果判断：180kDa处出现特异性条带，阴性对照（I型胶原提取物）无条带。
