产品介绍Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. They show extensive diversity in tissue distribution and in their subcellular localization. CA IX is a transmembrane protein and the only tumor-associated carbonic anhydrase isoenzyme known. It is expressed in all clear-cell renal cell carcinoma, but is not detected in normal kidney or most other normal tissues. It may be involved in cell proliferation and transformation. This gene was mapped to 17q21.2 by fluorescence in situ hybridization, however, radiation hybrid mapping localized it to 9p13-p12. [provided by RefSeq, Jul 2008]

靶点与功能

CA9（Carbonic Anhydrase IX）是缺氧诱导因子（HIF-1α）下游靶基因编码的跨膜酶（分子量约46kDa），核心功能与研究价值包括：

肿瘤微环境调控：通过催化CO₂+H₂O⇌H⁺+HCO₃⁻反应，维持肿瘤细胞内碱性pH（促进增殖）和细胞外酸性pH（促进侵袭），在肾细胞癌（RCC）、宫颈癌、乳腺癌等实体瘤中高表达（缺氧区域阳性率>80%）；

诊断标志物：肾透明细胞癌（ccRCC）中CA9阳性率>90%，是病理诊断的“金标准”之一（与VHL基因失活高度相关）；

治疗靶点：CA9抑制剂（如SLC-0111）联合PD-1抗体可逆转肿瘤酸性微环境，增强免疫治疗效果，其单抗偶联药物（ADC）已进入临床前研究。

实验操作关键要点

IHC-P肾细胞癌诊断：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），ccRCC组织细胞膜/细胞质呈棕黄色阳性，正常肾组织阴性；

阳性对照：已知CA9阳性的肾透明细胞癌组织切片；

判读标准：按阳性细胞比例计分（<10%阴性，10%-50%弱阳性，>50%强阳性），强阳性提示肿瘤缺氧程度高、预后差。

WB缺氧诱导验证：

样本处理：A498肾癌细胞（VHL缺陷型，组成型表达CA9）或Hela细胞（1% O₂缺氧培养48小时诱导CA9表达），RIPA裂解液（含1mM PMSF）冰上裂解30分钟；

抗体稀释：Proandybio MA824抗体1:800稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，46kDa处显清晰条带。

IF缺氧微环境共定位：

与HIF-1α抗体共染，CA9（绿色）在缺氧区域细胞膜表达，HIF-1α（红色）在细胞核表达，二者共定位区域为肿瘤缺氧核心区。
