产品介绍Calbindin is a calcium-binding protein belonging to the troponin C superfamily. It was originally described as a 27-kD protein induced by vitamin D in the duodenum of the chick. In the brain, its synthesis is independent of vitamin-D-derived hormones. Calbindin contains 4 active calcium-binding domains, and 2 modified domains that presumably have lost their calcium-binding capacity. 钙结合蛋白D28K（Calbindin D28K，CALB1）是EF手型钙结合蛋白（分子量约28kDa），核心功能与研究价值包括：

神经保护“钙缓冲剂”：在小脑浦肯野细胞、海马CA1区神经元中高表达，通过结合游离Ca²⁺（亲和力Kd≈0.5μM）避免钙超载诱导的凋亡，阿尔茨海默病模型中D28K⁺神经元存活率是阴性神经元的3倍；

钙转运“分子伴侣”：在肾脏近端小管、肠道吸收上皮细胞中，与钙通道TRPV5/6协同促进跨细胞钙转运，维生素D缺乏时表达量降低50%；

肿瘤标志物：在甲状腺乳头状癌中低表达（与BRAF突变负相关），可作为预后良好的独立指标（5年生存率提高28%）。

实验操作关键要点

WB神经元样本检测：

样本处理：大鼠小脑组织用含1mM CaCl₂的RIPA裂解液（维持钙结合活性）冰上裂解30分钟，12000g离心15分钟取上清；

抗体稀释：抗体1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温1小时，曝光时间30秒（28kDa处显清晰条带）；

阳性对照：小脑组织（强阳性）vs 肝脏组织（阴性）。

IHC-P神经定位：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），小脑浦肯野细胞胞质呈棕黄色颗粒状阳性，分子层树突可见“串珠样”分布；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟（减少非特异性钙结合）；

对比染色：与Parvalbumin（小清蛋白） 共染，可区分浦肯野细胞（D28K⁺）与篮状细胞（Parvalbumin⁺）。

IF钙信号共定位：

海马神经元爬片与Fluo-4 AM（钙探针） 共孵育，D28K（绿色）与高钙区域（红色）共定位系数>0.7，KCl刺激后共定位增强（钙结合活性上调）。
