产品介绍This gene encodes a calmodulin- and actin-binding protein that plays an essential role in the regulation of smooth muscle and nonmuscle contraction. The conserved domain of this protein possesses the binding activities to Ca(2+)-calmodulin, actin, tropomyosin, myosin, and phospholipids. This protein is a potent inhibitor of the actin-tropomyosin activated myosin MgATPase, and serves as a mediating factor for Ca(2+)-dependent inhibition of smooth muscle contraction. Alternative splicing of this gene results in multiple transcript variants encoding distinct isoforms.[provided by RefSeq, Jul 2008].

靶点与功能

Tubulin β是微管的主要结构蛋白（分子量约50kDa），作为内参的核心优势包括：

组成型表达：在所有真核细胞中稳定表达，不受细胞周期、营养状态或药物处理影响（HeLa细胞中Tubulin β mRNA半衰期>24小时）；

高保守性：不同物种间序列一致性>95%（人、小鼠、大鼠β-tubulin同源性达98%），可跨物种使用；

适用性广：适用于WB（最常用内参）、IHC、IF等多种实验，尤其适合总蛋白定量标准化（与GAPDH、β-actin并列为三大经典内参）。

实验操作关键要点

WB内参标准化：

样本处理：RIPA裂解液（含1mM PMSF）提取总蛋白，BCA定量后每孔上样20-30μg（确保线性范围内）；

抗体稀释：推荐1:2000（5%脱脂奶粉+TBST），推荐1:1000（5% BSA+TBST），4℃孵育过夜；

结果判读：50kDa处显清晰单一条带，目的蛋白灰度值需除以Tubulin β灰度值进行标准化（如目的蛋白/Tubulin β比值）。

IF细胞骨架共定位：

染色方案：与α-tubulin抗体共染（微管双标），β-tubulin（绿色）与α-tubulin（红色）在细胞内呈“网状微管结构”完全共定位，DAPI复染细胞核（蓝色）；

注意事项：固定用4%多聚甲醛（避免甲醇破坏微管结构），透化用0.1% Triton X-100（保持微管完整性）。

IHC组织表达验证：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复10分钟（石蜡切片），所有组织细胞胞质呈阳性（神经元轴突、肾小管上皮细胞信号最强）；

阴性对照：β-tubulin敲除细胞系（如HCT116 β-tubulin KO）或正常兔IgG（无信号）。
