产品介绍Calretinin is a calcium-binding protein which is abundant in auditory neurons. It belongs to the calbindin family. Calbindin 2 (calretinin), closely related to calbindin 1, is an intracellular calcium-binding protein belonging to the troponin C superfamily. Calbindin 1 is known to be involved in the vitamin-D-dependent calcium absorption through intestinal and renal epithelia, while the function of neuronal calbindin 1 and calbindin2 is poorly understood. The sequence of the calbindin 2 cDNA reveals an open reading frame of 271 codons coding for a protein of 31,520 Da, and shares 58% identical residues with human calbindin1.

实验操作关键要点

WB检测通用优化：

样本处理：使用含1mM CaCl₂的RIPA裂解液（维持钙结合活性），冰上裂解30分钟，12000g离心15分钟取上清；

抗体稀释：S100P单抗推荐1:1000（5% BSA+TBST），Calbindin-D28K单抗1:2000，4℃孵育过夜；

阳性对照：S100P用乳腺癌细胞系MDA-MB-231（高表达），Calbindin-D28K用大鼠小脑组织。

IHC-P组织定位：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15-20分钟，S100P在胃癌组织中胞质/胞膜阳性，Calbindin-D28K在小脑Purkinje细胞胞质强阳性；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性钙结合。

IF钙信号共定位：

与Fluo-4 AM（钙探针） 共孵育，激光共聚焦观察钙结合蛋白与Ca²⁺浓度变化的时空关联（如神经元激活时Calbindin-D28K与钙信号共定位增强）。
