产品介绍The protein encoded by this gene is a type II transmembrane glycoprotein and a common acute lymphocytic leukemia antigen that is an important cell surface marker in the diagnosis of human acute lymphocytic leukemia (ALL). The encoded protein is present on leukemic cells of pre-B phenotype, which represent 85% of cases of ALL. This protein is not restricted to leukemic cells, however, and is found on a variety of normal tissues. The protein is a neutral endopeptidase that cleaves peptides at the amino side of hydrophobic residues and inactivates several peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. CD10（Cluster of Differentiation 10），又称中性内肽酶（NEP） 或急性淋巴母细胞白血病共同抗原（CALLA），是一种II型跨膜锌依赖性金属蛋白酶（分子量约100kDa），核心功能与研究价值包括：

肿瘤标志物：在B细胞淋巴瘤中，CD10是生发中心细胞的特异性标志物（滤泡性淋巴瘤阳性率>90%，Burkitt淋巴瘤阳性率>85%），也是急性淋巴细胞白血病（ALL）的经典诊断指标（CALLA抗原）；

酶学功能：通过切割多种生物活性肽（如脑啡肽、P物质、血管紧张素），调控细胞增殖、分化和血管生成，在肾透明细胞癌、卵巢癌中高表达可促进肿瘤侵袭；

正常组织分布：表达于肾近端小管、小肠绒毛上皮、中性粒细胞等，可作为组织分化程度的参考指标。

实验操作关键要点

IHC-P淋巴瘤诊断（石蜡切片）：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟，CD10在滤泡性淋巴瘤生发中心呈细胞膜棕黄色阳性，套细胞淋巴瘤阴性（鉴别诊断关键）；

阳性对照：正常小肠黏膜绒毛上皮（细胞膜强阳性）；

临床判读：结合CD20、BCL-6共染，CD10⁺CD20⁺BCL-6⁺为典型生发中心B细胞表型。

WB肿瘤细胞系检测：

样本处理：Raji细胞（Burkitt淋巴瘤，CD10⁺）或A498肾癌细胞（CD10⁺），RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心15分钟取上清；

抗体稀释：按1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温1小时，100kDa处显清晰条带（肾癌细胞信号强于淋巴瘤细胞）。

IF免疫荧光共定位：

与CD20抗体共染（B细胞标志物），CD10（绿色）与CD20（红色）在生发中心B细胞共定位，共聚焦下可见细胞膜“环状双阳性”信号。
