产品介绍Aminopeptidase N is located in the small-intestinal and renal microvillar membrane, and also in other plasma membranes. In the small intestine aminopeptidase N plays a role in the final digestion of peptides generated from hydrolysis of proteins by gastric and pancreatic proteases. Its function in proximal tubular epithelial cells and other cell types is less clear. The large extracellular carboxyterminal domain contains a pentapeptide consensus sequence characteristic of members of the zinc-binding metalloproteinase superfamily. Sequence comparisons with known enzymes of this class showed that CD13 and aminopeptidase N are identical. The latter enzyme was thought to be involved in the metabolism of regulatory peptides by diverse cell types, including small intestinal and renal tubular epithelial cells, macrophages, granulocytes, and synaptic membranes from the CNS. This membrane-bound zinc metalloprotease is known to serve as a receptor for the HCoV-229E alphacoronavirus as well as other non-human coronaviruses. This gene has also been shown to promote angiogenesis, tumor growth, and metastasis and defects in this gene are associated with various types of leukemia and lymphoma. CD13（Aminopeptidase N, APN）是锌依赖性II型跨膜氨肽酶（分子量约150kDa），核心功能与研究价值包括：

肿瘤标志物：在急性髓系白血病（AML） 中阳性率>95%，是区分髓系（CD13⁺）与淋系白血病（CD13⁻）的关键指标；在肝细胞癌（HCC） 中高表达，与肿瘤分期和预后不良正相关（临床样本显示CD13⁺患者5年生存率降低37%）；

酶学功能：通过水解肽链N端氨基酸（如血管紧张素III→血管紧张素IV），调控血管生成、细胞侵袭和免疫抑制，在胰腺癌、卵巢癌中可促进肿瘤转移；

病毒受体：作为冠状病毒（如SARS-CoV-2）的潜在辅助受体，参与病毒入侵宿主细胞过程，是抗病毒研究的新兴靶点。

实验操作关键要点

WB膜蛋白检测：

样本处理：THP-1细胞（AML细胞系，CD13⁺）用膜蛋白提取试剂盒分离膜组分，RIPA裂解液（含1mM PMSF+10μM bestatin（APN抑制剂））冰上裂解30分钟；

抗体稀释：A按1:2000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温1小时，150kDa处显清晰条带（未处理组信号强于bestatin抑制组）。

IHC-P白血病骨髓切片：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），白血病细胞细胞膜呈棕黄色阳性，淋巴细胞阴性；

对照设置：正常骨髓组织（少量单核细胞弱阳性）、CD13敲除细胞系（阴性对照）；

显色控制：DAB显色5分钟，苏木素复染，阳性率=（CD13⁺细胞数/总细胞数）×100%，AML样本阳性率通常>80%。

Flow-Cyt免疫分型：

样本处理：外周血单个核细胞（PBMC）用PBS+2% FBS洗涤2次，CD13抗体（1:100稀释）4℃避光孵育30分钟，流式检测显示髓系细胞（CD13⁺CD33⁺）群比例，AML患者该群比例>20%（诊断阈值）。
