产品介绍The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK4 or CDK6, whose activity is required for cell cycle G1/S transition. This protein has been shown to interact with tumor suppressor protein Rb and the expression of this gene is regulated positively by Rb. Mutations, amplification and overexpression of this gene, which alters cell cycle progression, are observed frequently in a variety of tumors and may contribute to tumorigenesis. 靶点与功能

周期素D1（Cyclin D1，基因名CCND1）是G1期细胞周期调控的关键蛋白，通过与CDK4/CDK6结合形成活性复合物，磷酸化视网膜母细胞瘤蛋白（Rb），释放E2F转录因子，推动细胞从G1期进入S期。其异常高表达常见于乳腺癌、淋巴瘤、肺癌等多种肿瘤，是细胞过度增殖和癌变的重要标志物，也是肿瘤靶向治疗（如CDK4/6抑制剂）的疗效评估指标。

实验操作关键要点

WB优化（肿瘤细胞系）：

样本处理：使用含1mM NaF（磷酸酶抑制剂） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清，避免Cyclin D1去磷酸化影响检测；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳90分钟，确保36kDa条带与杂蛋白分离；

抗体孵育：一抗推荐1:2000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间5-10秒（避免信号过曝）。

IHC-P注意事项（乳腺癌组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强核内信号；

阳性对照：推荐使用乳腺浸润性导管癌组织（癌细胞核呈棕黄色强阳性，正常乳腺上皮弱阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性结合。

IP实验要点（蛋白互作研究）：

磁珠预处理：取20μL Protein A/G磁珠，用含蛋白酶抑制剂的PBS洗涤3次，加入5μg抗体4℃孵育2小时；

样本孵育：裂解液中加入1mg总蛋白，与抗体-磁珠复合物4℃旋转孵育过夜，洗涤5次后用SDS上样缓冲液洗脱，检测Cyclin D1与CDK4的相互作用。
