产品介绍Prostaglandin-endoperoxide synthase (PTGS), also known as cyclooxygenase, is the key enzyme in prostaglandin biosynthesis, and acts both as a dioxygenase and as a peroxidase. There are two isozymes of PTGS: a constitutive PTGS1 and an inducible PTGS2, which differ in their regulation of expression and tissue distribution. This gene encodes the inducible isozyme. It is regulated by specific stimulatory events, suggesting that it is responsible for the prostanoid biosynthesis involved in inflammation and mitogenesis. [provided by RefSeq, Feb 2009]

靶点与功能

COX-2（Cyclooxygenase-2）是诱导型环氧化酶，分子量约72kDa，在炎症刺激（如IL-1β、TNF-α）或肿瘤微环境中被激活，催化花生四烯酸转化为前列腺素（如PGE2）。其核心功能包括：

炎症调控：参与疼痛、发热等炎症反应，是非甾体抗炎药（NSAIDs）的主要靶点；

肿瘤关联：在结直肠癌、胃癌、乳腺癌等肿瘤中高表达，通过促进血管生成、抑制凋亡推动肿瘤进展；

免疫调节：调控肿瘤相关巨噬细胞（TAM）极化，促进免疫抑制性微环境形成。

实验操作关键要点

WB优化（炎症细胞模型）：

样本处理：LPS（1μg/mL）刺激RAW264.7细胞6小时后，用含磷酸酶抑制剂的RIPA裂解液冰上裂解20分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约72kDa；

抗体孵育：一抗1:2000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（结直肠癌组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性定位：肿瘤细胞胞质呈棕黄色颗粒状阳性，癌旁炎症区域信号增强；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性结合。
