产品介绍The cyclophilins are a conserved class of proteins that bind the immunosuppressive drug cyclosporin A (CsA) with high affinity. CsA blocks helper T-cell activation at a step between T-cell receptor stimulation and the transcriptional activation of cytokine genes. Cyclophilins from many species possess peptidyl-prolyl cis-trans isomerase (PPIase) activity that is blocked by CsA and therefore may be relevant in CsA-mediated immunosuppression.

PPIB（Peptidylprolyl Isomerase B）是亲环蛋白家族成员，分子量约21kDa，广泛表达于真核生物各组织和细胞中，主要定位于内质网。其核心功能包括：

内参特性：表达稳定（不受常见实验处理如药物、缺氧影响），适合作为WB、IHC等实验的内参对照，用于校正上样量差异；

生理功能：催化肽基脯氨酸顺反异构化，参与蛋白质折叠与成熟，在免疫调节、炎症反应中起辅助作用。

实验操作关键要点

WB内参应用（细胞/组织样本）：

样本处理：RIPA裂解液（含蛋白酶抑制剂）冰上裂解30分钟，12000g离心取上清，BCA法准确定量蛋白浓度；

上样量控制：推荐上样20-30μg总蛋白，确保目的蛋白与内参条带在同一线性范围内；

抗体孵育：一抗1:3000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光时间5-15秒（避免过曝）。

内参有效性验证：

稳定性检测：对比不同处理组（如药物处理、细胞分化）的PPIB条带灰度值，变异系数应<10%；

条带单一性：WB中应出现单一清晰条带（21kDa左右），无明显杂带或二聚体；

组织特异性：避免在精子、成熟红细胞等特殊组织中使用（PPIB表达较低）。
