产品介绍This gene encodes a member of the keratin family, the most diverse group of intermediate filaments. This gene product, a type I keratin, is usually found as a heterotetramer with two keratin 5 molecules, a type II keratin. Together they form the cytoskeleton of epithelial cells. Mutations in the genes for these keratins are associated with epidermolysis bullosa simplex. At least one pseudogene has been identified at 17p12-p11. [provided by RefSeq].

靶点与功能

细胞角蛋白14（Cytokeratin 14, CK14）是I型酸性中间丝蛋白，分子量约50kDa，由KRT14基因编码。其核心功能与研究价值包括：

组织特异性：主要表达于复层鳞状上皮的基底细胞（如皮肤表皮、食管上皮、宫颈上皮），是上皮干细胞的标志物；

肿瘤诊断：在鳞状细胞癌（如皮肤鳞癌、宫颈鳞癌、头颈部鳞癌）中高表达，阳性率>90%，可用于与腺癌、小细胞癌的鉴别；

皮肤生物学：参与皮肤屏障维持和伤口愈合，其突变与大疱性表皮松解症等遗传性皮肤病密切相关。

实验操作关键要点

IHC-P鳞状细胞癌诊断：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞质呈弥漫性棕黄色阳性，鳞癌巢周边基底样细胞表达更强；

抗体稀释：推荐1:200稀释（5% BSA+TBST），4℃孵育过夜，HRP聚合物二抗室温孵育30分钟。

WB验证（皮肤组织样本）：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1.5小时）；

结果判断：50kDa处出现单一特异性条带，阴性对照（如腺癌组织）无条带。
