产品介绍The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. 靶点与功能

细胞角蛋白19（Cytokeratin 19, CK19）是I型酸性角蛋白，分子量约40kDa，由KRT19基因编码。其核心功能与研究价值包括：

组织特异性：主要表达于单层上皮细胞（如乳腺导管、胆管、肺泡上皮），是上皮细胞分化的关键标志物；

肿瘤诊断：在腺癌（如乳腺癌、肺癌、胰腺癌）中高表达，阳性率>85%，与CK7联合检测可提高诊断特异性；

临床应用：循环肿瘤细胞（CTC）检测的重要标志物（如CellSearch系统），可用于癌症转移监测和预后评估。

实验操作关键要点

IHC-P腺癌诊断（乳腺癌样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复20分钟（95℃），冷却后用3% H₂O₂封闭10分钟；

阳性定位：肿瘤细胞胞质呈弥漫性棕黄色阳性，腺管结构周边细胞表达更强；

对照设置：必须包含正常乳腺组织（阳性对照）和鳞癌组织（阴性对照）。

WB验证（细胞裂解液）：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶（分离小分子量蛋白），恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（200mA恒流1小时）；

抗体稀释：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光时间15-30秒。
