产品介绍The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. 研究意义：作为乳腺干细胞、肝干细胞的表面标志物，可用于干细胞分离鉴定及上皮-间质转化（EMT）机制研究。

实验操作关键要点

IHC-P优化（肺癌组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强胞质信号；

阳性定位：肿瘤细胞胞质呈细丝状或弥漫性棕黄色阳性，鳞癌表达强度高于腺癌；

对照设置：必须包含正常支气管上皮（阳性对照）和间叶组织（阴性对照）。

WB检测注意事项：

样本处理：RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心取上清；若检测CYFRA21-1片段，需添加蛋白酶抑制剂（如EDTA）；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约44kDa；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
