产品介绍This gene encodes a carcinoma-associated antigen and is a member of a family that includes at least two type I membrane proteins. This antigen is expressed on most normal epithelial cells and gastrointestinal carcinomas and functions as a homotypic calcium-independent cell adhesion molecule. The antigen is being used as a target for immunotherapy treatment of human carcinomas. Mutations in this gene result in congenital tufting enteropathy. [provided by RefSeq, Dec 2008]

靶点与功能

CD326（EpCAM）是一种跨膜糖蛋白，分子量约40kDa，含265个氨基酸（胞外域242aa、跨膜域23aa、胞内域26aa），核心功能包括：

生理作用：介导上皮细胞间粘附，参与细胞增殖与分化信号转导；

临床价值：上皮性肿瘤的关键标志物（乳腺癌、结肠癌、前列腺癌等过表达），与肿瘤侵袭转移、预后不良正相关，也是肿瘤干细胞（CSCs）的表面标志物；

研究意义：作为CAR-T细胞治疗靶点（如针对EpCAM的免疫疗法），是肿瘤免疫治疗机制研究的重要工具。

实验操作关键要点

IHC-P组织染色（肿瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强胞膜信号；

阳性定位：上皮细胞细胞膜呈棕黄色线性阳性，癌组织中表达强度高于正常组织；

对照设置：需包含正常上皮组织（弱阳性）和间叶组织（阴性对照）。

WB验证（结肠癌细胞裂解液）：

样本制备：使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

流式细胞术（肿瘤干细胞分选）：

样本处理：单细胞悬液用PBS洗涤2次，0.5% BSA封闭30分钟；

抗体标记：抗体1:100稀释，4℃避光孵育30分钟，流式缓冲液洗涤后上机检测；

阳性判定：CD326⁺细胞群可通过荧光强度阈值与阴性对照（同型抗体）区分。
