产品介绍Estrogen and progesterone receptor are members of a family of transcription factors that are regulated by the binding of their cognate ligands. The interaction of hormone-bound estrogen receptors with estrogen responsive elements(EREs) alters transcription of ERE-containing genes. The carboxy terminal region of the estrgen receptor contains the ligand binding domain, the amino terminus serves as the transactivation domain, and the DNA binding domain is centrally located. Two forms of estrogen receptor have been identified, ER Alpha and ER Beta. ER Alpha and ER Beta have been shown to be differentially activated by various ligands. The biological response to progesterone is mediated by two distinct forms of the human progesterone receptor (hPR-A and hPR-B), which arise from alternative splicing. In most cells, hPR-B functions as a transcriptional activator of progesterone-responsive gene, whereas hPR-A function as a transcriptional inhibitor of all steroid hormone receptors.

抗体特性

宿主来源：兔多克隆抗体。

交叉反应：与人（Human）、小鼠（Mouse）、大鼠（Rat）、狗（Dog）等物种的AANAT蛋白具有反应性。

分子量：预测约23kDa（需通过Western Blot验证）。

免疫原：KLH偶联的人源AANAT蛋白片段（161-207aa）。

纯化方式：Protein A亲和纯化。

保存条件：

液体抗体：含0.01M PBS（pH 7.4）、10mg/mL BSA及0.1%叠氮化钠，建议-20℃保存，避免反复冻融。

冻干粉：室温下稳定至少1个月，-20℃可保存1年以上。

实验操作关键要点

IHC-P乳腺癌组织染色（临床诊断）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 高压修复2分钟（121℃），冷却后用3% H₂O₂阻断内源性过氧化物酶10分钟；

阳性定位：ERα阳性乳腺癌细胞细胞核强阳性（核染色比例≥1%为阳性），正常乳腺上皮细胞呈弱阳性；

染色流程：一抗推荐1:150稀释（5% BSA+TBST），4℃孵育过夜，EnVision二抗室温孵育30分钟，DAB显色5分钟，苏木素复染细胞核。

ChIP实验（ERα靶基因验证）：

样本制备：MCF-7细胞用10nM E2刺激45分钟，1%甲醛交联10分钟，甘氨酸终止后超声破碎（片段200-500bp）；

抗体孵育：抗体5μg与Protein A/G磁珠4℃孵育2小时，加入超声上清4℃旋转孵育过夜；

洗脱与检测：解交联后提取DNA，qPCR检测GREB1基因启动子ERE区域富集倍数（E2处理组较对照组≥5倍）。

WB检测注意事项：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液，避免ERα降解；推荐MCF-7细胞（ERα+）为阳性对照，MDA-MB-231（ERα-）为阴性对照；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光时间30秒。
