产品介绍This gene encodes fibronectin, a glycoprotein present in a soluble dimeric form in plasma, and in a dimeric or multimeric form at the cell surface and in extracellular matrix. Fibronectin is involved in cell adhesion and migration processes including embryogenesis, wound healing, blood coagulation, host defense, and metastasis. The gene has three regions subject to alternative splicing, with the potential to produce 20 different transcript variants. However, the full-length nature of some variants has not been determined. 靶点与功能

纤维连接蛋白（Fibronectin，FN）是细胞外基质中的多功能糖蛋白，分子量约250kDa（二聚体形式），由两条相似的亚基通过二硫键连接。其核心功能包括：

生理作用：介导细胞粘附（通过RGD序列结合整合素受体）、促进细胞迁移与分化；

临床价值：肿瘤组织中FN表达异常（如FN1基因高表达）与肿瘤侵袭、血管生成、预后不良正相关，是癌症转移风险评估的潜在标志物；

研究意义：参与EMT（上皮-间质转化）过程，是肿瘤微环境重塑机制研究的关键分子。

实验操作关键要点

IHC-P组织染色（肿瘤微环境样本）：

抗原修复：采用胃蛋白酶消化法（0.4%胃蛋白酶+0.1M HCl，37℃孵育20分钟），增强ECM信号暴露；

阳性定位：肿瘤间质区纤维状或网状棕黄色阳性，癌巢周围表达增强；

对照设置：需包含正常组织（如肝脏）（弱阳性）和FN敲除小鼠组织（阴性对照）。

WB验证（细胞培养上清/组织裂解液）：

样本处理：细胞上清需经40%硫酸铵沉淀（4℃静置2小时），离心后用PBS复溶；

电泳条件：7.5% SDS-PAGE凝胶（FN分子量较大），恒压80V浓缩胶、100V分离胶，转膜选用0.22μm PVDF膜（300mA恒流2小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IF共定位实验（细胞爬片）：

固定条件：4%多聚甲醛室温固定15分钟，0.1% Triton X-100透化10分钟；

荧光标记：与整合素β1抗体共染，激光共聚焦显微镜下观察“FN-整合素”复合物的膜定位。
