产品介绍The protein encoded by this gene is a member of the forkhead/winged-helix family of transcriptional regulators. Defects in this gene are the cause of immunodeficiency polyendocrinopathy, enteropathy, X-linked syndrome (IPEX), also known as X-linked autoimmunity-immunodeficiency syndrome. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008].

靶点与功能

FOXP3是叉头框转录因子家族成员，分子量约47kDa，主要表达于Treg细胞，通过调控IL-2、CTLA-4等基因表达维持免疫抑制功能。其核心功能与疾病关联包括：

免疫调节：Treg细胞中FOXP3持续表达是免疫耐受的分子基础，缺失会导致严重自身免疫病（如IPEX综合征）；

肿瘤微环境：肿瘤浸润Treg细胞高表达FOXP3可抑制抗肿瘤免疫，与胃癌、肺癌的预后不良相关；

治疗靶点：通过抗体检测FOXP3⁺ Treg比例可评估免疫治疗响应，是免疫检查点抑制剂疗效预测的潜在标志物。

实验操作关键要点

IHC-P优化（淋巴结组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强核定位信号；

阳性定位：Treg细胞细胞核呈棕黄色阳性，主要分布于淋巴滤泡间区及肿瘤间质；

封闭处理：5%驴血清+0.1% Triton X-100室温封闭30分钟，减少非特异性核染色。

WB检测注意事项：

样本处理：使用含1mM Na₃VO₄（磷酸酶抑制剂） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清（保留转录因子活性）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约47kDa；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
