产品介绍The protein encoded by this gene has been termed a 'growth' or 'plasticity' protein because it is expressed at high levels in neuronal growth cones during development and axonal regeneration. This protein is considered a crucial component of an effective regenerative response in the nervous system. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

靶点与功能

GAP43是轴突生长锥标志性蛋白，分子量约43kDa，在神经元发育、轴突再生及突触可塑性中高表达。其核心功能与研究价值包括：

神经再生：轴突生长锥中GAP43通过Ser41/Ser96磷酸化调控细胞骨架重组，是神经损伤修复的关键分子标志物；

突触可塑性：在海马、皮层等脑区高表达，参与学习记忆形成，与阿尔茨海默病、帕金森病的神经退行性变密切相关；

发育研究：胚胎期神经元轴突延伸阶段高表达，可用于神经回路构建的动态追踪。

实验操作关键要点

IHC-P优化（脑组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），增强轴突信号；

阳性定位：生长锥及突触前膜呈棕黄色颗粒状阳性，再生神经纤维中表达显著上调；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭30分钟，减少非特异性背景。

WB检测注意事项：

样本处理：使用含1mM Na₃VO₄（磷酸酶抑制剂） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清（保留磷酸化状态）；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约43kDa；

抗体孵育：小鼠单抗1:8000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗小鼠）1:5000室温孵育1小时。
