产品介绍Members of the GATA family share a conserved zinc finger DNA-binding domain and are capable of binding the WGATAR consensus sequence. GATA-1 is erythroid-specific and is responsible for the regulated transcription of erythroid genes. It is an essential component in the generation of the erythroid lineage. GATA-2 is expressed in embryonic brain and liver, HeLa and endothelial cells, as well as in erythroid cells. Studies with a modified GATA consensus sequence, AGATCTTA, have shown that GATA-2 and GATA-3 recognize this mutated consensus while GATA-1 has poor recognition of this sequence. This indicates broader regulatory capabilities of GATA-2 and GATA-3 than GATA-1. GATA-3 is highly expressed in T lymphocytes. GATA-4, GATA-5 and GATA-6 comprise a subfamily of transcription factors. Both GATA-4 and GATA-6 are found in heart, pancreas and ovary; lung and liver tissues exhibit GATA-6, but not GATA-4 expression. GATA-5 expression has been observed in differentiated heart and gut tissues and is present throughout the course of development in the heart. Although expression patterns of the various GATA transcription factors may overlap, it is not yet apparent how the GATA factors are able to discriminate in binding their appropriate target sites.

靶点与功能

GATA3是锌指家族转录因子，定位于10p14-p15染色体，通过结合W GATAR（W=A/T，R=A/G） consensus序列调控基因表达。其核心功能包括：

生理作用：参与T细胞分化（Th2亚群极化）、乳腺/泌尿生殖道上皮发育；

临床价值：乳腺癌（尤其是luminal型）的特异性标志物（阳性率约70-80%），与ER/PR联合检测可提高分型准确性；甲状腺乳头状癌、膀胱尿路上皮癌中也呈高表达；

研究意义：通过ChIP-seq鉴定GATA3靶基因，揭示其在肿瘤微环境重塑（如EMT抑制）中的作用。

实验操作关键要点

IHC-P组织染色（乳腺癌样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），避免EDTA缓冲液影响核定位信号；

阳性定位：正常乳腺上皮细胞细胞核呈棕黄色阳性，癌组织中表达强度与肿瘤分化程度正相关；

对照设置：必须包含正常乳腺组织（阳性对照）和肺腺癌组织（阴性对照，排除交叉反应）。

WB验证（细胞裂解液）：

样本处理：MCF-7细胞（高表达GATA3）用含蛋白酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（250mA恒流1小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
