靶点与功能

GCDFP-15（Gross Cystic Disease Fluid Protein-15）是分泌型糖蛋白，分子量约13.5kDa，主要表达于乳腺小叶和导管上皮细胞，通过与肌动蛋白结合参与细胞分泌功能。其核心功能与研究价值包括：

肿瘤诊断：在乳腺浸润性导管癌中高表达，是鉴别乳腺癌与其他腺癌（如肺癌、卵巢癌）的特异性标志物；

预后评估：乳腺癌组织中GCDFP-15阳性提示激素受体依赖性，与较好的化疗响应相关；

病理鉴别：在涎腺肿瘤、汗腺癌中也可表达，可辅助判断肿瘤组织来源。

实验操作关键要点

IHC-P优化（乳腺癌石蜡切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强腺上皮信号；

阳性定位：乳腺导管上皮细胞胞质呈棕黄色颗粒状阳性，癌组织中表达强度与肿瘤分化程度正相关；

对照设置：必须包含正常乳腺组织（阳性对照）和肺腺癌组织（阴性对照，排除交叉反应）。

WB检测注意事项：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心取上清；若检测分泌型蛋白，需收集细胞培养液上清；

电泳条件：15% SDS-PAGE凝胶（因分子量较小），恒压80V浓缩胶、120V分离胶，目标条带约13.5kDa；

抗体孵育：兔单抗1:800稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
