产品介绍This gene encodes one of the major intermediate filament proteins of mature astrocytes. It is used as a marker to distinguish astrocytes from other glial cells during development. Mutations in this gene cause Alexander disease, a rare disorder of astrocytes in the central nervous system. Alternative splicing results in multiple transcript variants encoding distinct isoforms. GFAP是星形胶质细胞特异性中间丝蛋白，分子量约50kDa，参与维持细胞骨架结构、血脑屏障完整性及神经修复。其核心功能与研究价值包括：

细胞标记：作为星形胶质细胞的金标准标志物，用于区分神经元（如NeuN标记）与胶质细胞；

神经损伤：脑缺血、创伤后GFAP表达上调，是胶质瘢痕形成的关键指标；

疾病关联：在胶质母细胞瘤、多发性硬化中异常高表达，可辅助肿瘤分级与预后评估。

实验操作关键要点

IHC-P优化（脑组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），避免EDTA缓冲液影响信号；

阳性定位：星形胶质细胞胞质及突起呈棕黄色丝状阳性，灰质中呈放射状分布，白质中沿神经纤维排列；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭30分钟，减少非特异性结合。

WB检测注意事项：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心取上清（星形胶质细胞纯度建议>80%）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，目标条带约50kDa；

抗体孵育：单抗1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。
