产品介绍This gene encodes a receptor for the Fc portion of immunoglobulin G, and it is involved in the removal of antigen-antibody complexes from the circulation, as well as other responses, including antibody dependent cellular mediated cytotoxicity and antibody dependent enhancement of virus infections. This gene (FCGR3A) is highly similar to another nearby gene (FCGR3B) located on chromosome 1. The receptor encoded by this gene is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, whereas FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. Mutations in this gene are associated with immunodeficiency 20, and have been linked to susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia. 抗体特性

实验操作关键要点

WB检测（NK细胞裂解液）：

样本制备：使用含1mM PMSF和磷酸酶抑制剂的RIPA裂解液冰上裂解NK-92细胞30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（300mA恒流1.5小时）；

抗体孵育：一抗1:1500稀释（5% BSA+TBST），4℃孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间45秒（避免信号过强）。

IHC-P组织染色（淋巴瘤样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复10分钟（石蜡切片），自然冷却至室温；

阳性定位：NK细胞、巨噬细胞胞膜呈棕黄色阳性，肿瘤微环境中浸润的NK细胞数量与预后正相关；

对照设置：必须包含扁桃体组织（阳性对照，NK细胞富集区显色）和FCGR3A敲除细胞系（阴性对照）。

流式细胞术（NK细胞表面标记）：

样本处理：外周血NK细胞（CD3⁻CD56⁺）用含2% FBS的PBS洗涤2次，4℃封闭30分钟；

抗体稀释：抗体用FACS缓冲液按1:100稀释，4℃避光孵育1小时，流式细胞仪检测时以同型抗体为阴性对照；

结果分析：FCGR3A⁺细胞比例正常参考值为NK细胞的85%-95%，比例降低提示免疫缺陷或抗体治疗耐药风险。
