产品介绍This gene encodes a member of the immunoglobulin gene superfamily. Expression of this cell surface protein is restricted to B cell lymphocytes. This protein is a reliable marker for pre-B cells but its expression diminishes during terminal B cell differentiation in antibody secreting plasma cells. The protein has two N-terminal extracellular Ig-like domains separated by a non-Ig-like domain, a hydrophobic transmembrane domain, and a large C-terminal cytoplasmic domain. This protein forms a complex with several membrane proteins including complement receptor type 2 (CD21) and tetraspanin (CD81) and this complex reduces the threshold for antigen-initiated B cell activation. Activation of this B-cell antigen receptor complex activates the phosphatidylinositol 3-kinase signalling pathway and the subsequent release of intracellular stores of calcium ions. This protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of lymphoblastic leukemia. Mutations in this gene are associated with the disease common variable immunodeficiency 3 (CVID3) which results in a failure of B-cell differentiation and impaired secretion of immunoglobulins. CVID3 is characterized by hypogammaglobulinemia, an inability to mount an antibody response to antigen, and recurrent bacterial infections. 实验操作关键要点

流式细胞术（外周血B细胞检测）：

设门策略：先圈定淋巴细胞群（SSC-A/FSG-A），再通过CD19+CD20+双阳性确定B细胞亚群（正常参考值：外周血B细胞占淋巴细胞10%-20%）。

IHC-P组织染色（淋巴瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），冷却至室温；

阳性定位：B淋巴细胞细胞膜呈棕黄色线性阳性，在滤泡性淋巴瘤中呈结节状分布，弥漫大B细胞淋巴瘤中弥漫阳性；

对照设置：必须包含正常扁桃体组织（生发中心B细胞强阳性）和T细胞淋巴瘤组织（阴性对照）。

WB验证（Raji细胞裂解液）：

样本制备：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（300mA恒流2小时）；

抗体孵育：兔单抗1:2000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间60秒。
