产品介绍This gene encodes a member of the immunoglobulin gene superfamily. Expression of this cell surface protein is restricted to B cell lymphocytes. This protein is a reliable marker for pre-B cells but its expression diminishes during terminal B cell differentiation in antibody secreting plasma cells. The protein has two N-terminal extracellular Ig-like domains separated by a non-Ig-like domain, a hydrophobic transmembrane domain, and a large C-terminal cytoplasmic domain. This protein forms a complex with several membrane proteins including complement receptor type 2 (CD21) and tetraspanin (CD81) and this complex reduces the threshold for antigen-initiated B cell activation. Activation of this B-cell antigen receptor complex activates the phosphatidylinositol 3-kinase signalling pathway and the subsequent release of intracellular stores of calcium ions. This protein is a target of chimeric antigen receptor (CAR) T-cells used in the treatment of lymphoblastic leukemia. Mutations in this gene are associated with the disease common variable immunodeficiency 3 (CVID3) which results in a failure of B-cell differentiation and impaired secretion of immunoglobulins. CVID3 is characterized by hypogammaglobulinemia, an inability to mount an antibody response to antigen, and recurrent bacterial infections. 靶点与功能

CD19（B淋巴细胞表面抗原）是B细胞谱系的特异性标志物，属于Ig超家族跨膜蛋白，从早期B细胞前体到成熟B细胞阶段持续表达，在浆细胞分化后期消失。其核心功能包括：

B细胞活化调控：与BCR（B细胞受体）形成共受体复合物，降低B细胞活化阈值，促进B细胞增殖与抗体产生；

肿瘤治疗靶标：在B细胞急性淋巴细胞白血病（B-ALL）、弥漫性大B细胞淋巴瘤（DLBCL）等恶性肿瘤中高表达，是CAR-T疗法、单克隆抗体药物的核心靶点（如已获批的坦昔妥单抗、伊奈利珠单抗）。

实验操作关键要点

WB优化（B细胞系样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，推荐使用Raji细胞（B淋巴瘤细胞系） 作为阳性对照，K562细胞（非B细胞系）作为阴性对照；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳90分钟确保95kDa条带分离；

抗体孵育：一抗推荐1:1500稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒。

Flow-Cyt检测（外周血B细胞）：

样本制备：EDTA抗凝全血经红细胞裂解液处理后，用PBS洗涤2次，调整细胞浓度至1×10⁶ cells/mL；

抗体孵育：CD19抗体（如Alexa Fluor 488标记）按1:100稀释，4℃避光孵育30分钟，流式细胞仪检测B细胞（CD19⁺CD20⁺）比例；

对照设置：需设置同型对照（兔IgG）排除非特异性结合。

IHC-P注意事项（淋巴瘤组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强膜蛋白信号；

阳性定位：B细胞淋巴瘤组织中细胞膜呈棕黄色阳性，正常淋巴结组织中B细胞区（生发中心）阳性；

封闭处理：5%驴血清+0.1% Triton X-100室温封闭45分钟，降低背景染色。
