产品介绍Glycophorins A (GYPA) and B (GYPB) are major sialoglycoproteins of the human erythrocyte membrane which bear the antigenic determinants for the MN and Ss blood groups. In addition to the M or N and S or s antigens that commonly occur in all populations, about 40 related variant phenotypes have been identified. These variants include all the variants of the Miltenberger complex and several isoforms of Sta, as well as Dantu, Sat, He, Mg, and deletion variants Ena, S-s-U- and Mk. CD235a（糖蛋白A，Glycophorin A）是红细胞膜表面最丰富的糖蛋白，属于糖基磷脂酰肌醇（GPI）锚定蛋白，分子量约32kDa，主要功能包括：

结构作用：维持红细胞膜稳定性，参与膜骨架与脂质双层的连接；

血型抗原：携带MN血型系统的抗原决定簇（M抗原：Ser1、Gly5；N抗原：Leu1、Glu5），其基因多态性（如GYPA*M/*N）决定MN血型表型；

病原体受体：作为恶性疟原虫EBA-175蛋白和流感病毒的受体，在寄生虫感染机制研究中至关重要；

临床价值：在遗传性球形红细胞增多症中表达降低，在胎儿有核红细胞检测（产前诊断）中作为特异性标志物。

实验操作关键要点

IHC-F检测胎儿有核红细胞：

样本处理：孕妇外周血经密度梯度离心（如Ficoll-Paque）分离单个核细胞，涂片后4%多聚甲醛固定15分钟；

染色步骤：一抗1:200稀释（5% BSA+PBS），4℃孵育过夜，Alexa Fluor 488标记二抗（1:500）室温孵育1小时，DAPI复染细胞核；

结果判断：有核红细胞呈胞膜绿色荧光+核蓝色荧光，需排除CD45+白细胞（双标染色区分）。

FCM分析红细胞纯度：

抗体孵育：1×10⁶红细胞用0.1% BSA-PBS洗涤2次，加入抗体（1:100稀释）4℃避光孵育30分钟；

对照设置：需包含正常红细胞（阳性对照）和CD235a敲除细胞（阴性对照，如K562细胞）；

流式参数：激发波长488nm，发射波长525nm（FITC标记），通过FSC/SSC圈门排除碎片。

WB验证（红细胞膜蛋白提取）：

样本制备：取1mL全血，1500g离心5分钟收集红细胞，用低渗缓冲液（5mM磷酸钠，pH7.4） 裂解红细胞膜，12000g离心20分钟取沉淀；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（200mA恒流1.5小时）。
