产品介绍The DPP4 gene encodes dipeptidyl peptidase 4, which is identical to adenosine deaminase complexing protein-2, and to the T-cell activation antigen CD26. It is an intrinsic type II transmembrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. Dipeptidyl peptidase 4 is highly involved in glucose and insulin metabolism, as well as in immune regulation. This protein was shown to be a functional receptor for Middle East respiratory syndrome coronavirus (MERS-CoV), and protein modeling suggests that it may play a similar role with SARS-CoV-2, the virus responsible for COVID-19. 靶点与功能

CD26（二肽基肽酶IV，DPP4）是一种跨膜丝氨酸蛋白酶，分子量约110kDa（同源二聚体形式），主要表达于活化T细胞、上皮细胞及内皮细胞表面。其核心功能包括：

酶活性作用：特异性切割肽链N端第2位脯氨酸残基，灭活GLP-1、GIP等肠促胰岛素，参与血糖调控（DPP4抑制剂是2型糖尿病治疗药物）；

免疫调节：作为T细胞共刺激分子，通过与腺苷脱氨酶（ADA）结合增强T细胞活化，在关节炎、多发性硬化等自身免疫病中表达异常；

肿瘤微环境：在肺癌、结直肠癌中高表达，通过促进肿瘤细胞侵袭和血管生成参与疾病进展，是潜在的治疗靶点。

实验操作关键要点

WB检测（T细胞活化模型）：

样本处理：Jurkat细胞经PHA（5μg/mL） 刺激48小时，RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（300mA恒流2小时）；

抗体孵育：一抗1:2000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔/小鼠）1:5000室温孵育1小时，曝光时间60-90秒。

FCM分析（外周血T细胞）：

抗体组合：PerCP标记CD26抗体（1μg/100μL细胞）与APC-CD3、PE-CD4抗体共染，4℃避光30分钟；

结果判断：正常人群CD4⁺T细胞中CD26⁺比例约60%-80%，活化后表达上调（可通过CD69共标验证）。

IHC-F（冷冻组织切片）：

抗原修复：4%多聚甲醛固定15分钟后，0.1% Triton X-100透化10分钟，5%驴血清封闭30分钟；

阳性定位：小肠上皮细胞刷状缘、活化T细胞细胞膜呈绿色荧光（Alexa Fluor 488标记）。
