产品介绍The protein encoded by this gene is a non-lineage-restricted, type II transmembrane glycoprotein that synthesizes and hydrolyzes cyclic adenosine 5'-diphosphate-ribose, an intracellular calcium ion mobilizing messenger. The release of soluble protein and the ability of membrane-bound protein to become internalized indicate both extracellular and intracellular functions for the protein. This protein has an N-terminal cytoplasmic tail, a single membrane-spanning domain, and a C-terminal extracellular region with four N-glycosylation sites. Crystal structure analysis demonstrates that the functional molecule is a dimer, with the central portion containing the catalytic site. It is used as a prognostic marker for patients with chronic lymphocytic leukemia. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Sep 2015]

靶点与功能

CD38（Cluster of Differentiation 38）是II型跨膜糖蛋白（分子量约45kDa），核心功能与研究价值包括：

肿瘤治疗靶点：在多发性骨髓瘤（MM） 浆细胞中高表达（阳性率>95%），通过ADCC、ADCP和CDC效应清除肿瘤细胞，是**获批治疗MM的CD38单抗（达雷妥尤单抗）的作用靶点；

免疫调节作用：在NK细胞、T细胞、浆细胞表面表达，通过水解NAD+生成cADPR调节钙信号，参与B细胞分化和巨噬细胞吞噬功能（如ITP中抑制巨噬细胞吞噬血小板）；

自免疾病应用：清除CD38+浆细胞和活化免疫细胞，降低自身抗体（如抗ds-DNA）水平，在系统性红斑狼疮（SLE）、原发性免疫性血小板减少症（ITP）中显示疗效。

实验操作关键要点

WB检测MM细胞系表达：

样本处理：U266细胞用RIPA裂解液（含1mM PMSF） 冰上裂解30分钟，12000g离心15分钟取上清，45kDa处显清晰条带（阴性对照为Raji细胞，CD38低表达）；

抗体稀释：按1:200稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温1小时。

IHC-P浆细胞定位：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），MM骨髓组织中浆细胞细胞膜呈强阳性，正常骨髓浆细胞弱阳性；

封闭处理：3% H₂O₂室温10分钟，5% BSA封闭30分钟，抗体1:100稀释，4℃孵育过夜。

FCM免疫细胞分型：

抗体组合：CD38-PE（1:50）+ CD138-FITC（1:50）共染，MM细胞表型为CD38⁺⁺CD138⁺，NK细胞为CD38⁺CD56⁺。
