产品介绍The protein encoded by this gene is a non-lineage-restricted, type II transmembrane glycoprotein that synthesizes and hydrolyzes cyclic adenosine 5'-diphosphate-ribose, an intracellular calcium ion mobilizing messenger. The release of soluble protein and the ability of membrane-bound protein to become internalized indicate both extracellular and intracellular functions for the protein. This protein has an N-terminal cytoplasmic tail, a single membrane-spanning domain, and a C-terminal extracellular region with four N-glycosylation sites. Crystal structure analysis demonstrates that the functional molecule is a dimer, with the central portion containing the catalytic site. It is used as a prognostic marker for patients with chronic lymphocytic leukemia. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Sep 2015]

实验操作关键要点

流式细胞术（MM细胞分选）：

样本处理：骨髓穿刺液经Ficoll密度梯度离心获取单个核细胞，CD38抗体（PE标记）1:50稀释，4℃避光孵育30分钟，流式分选CD38⁺⁺CD138⁺ MM细胞（典型表型）；

注意事项：避免使用EDTA抗凝管（可能影响抗原构象），样本离体后2小时内染色。

IHC-P骨髓活检诊断：

抗原修复：EDTA缓冲液（pH9.0）高压修复15分钟（石蜡切片），MM细胞细胞膜呈棕黄色强阳性，正常浆细胞弱阳性，淋巴细胞阴性；

抗体稀释：Proteintech 66286-1-Ig按1:200稀释（5% BSA+TBST），4℃孵育过夜，DAB显色5分钟（控制背景）。

WB检测酶活性调控：

样本处理：细胞经达雷妥尤单抗（10μg/ml）处理48小时，RIPA裂解液（含1mM PMSF）冰上裂解，12000g离心15分钟取上清；

结果分析：45kDa条带灰度值较对照组降低40%（提示CD38内吞降解），同步检测p-ERK1/2（下游信号抑制）。
