产品介绍The protein encoded by this gene is the CD3-epsilon polypeptide, which together with CD3-gamma, -delta and -zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T-cell receptor-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development. Defects in this gene cause immunodeficiency. This gene has also been linked to a susceptibility to type I diabetes in women. [provided by RefSeq, Jul 2008].

靶点与功能

CD3是由ε、γ、δ、ζ、η链组成的跨膜蛋白复合物，其中CD3-ε链（分子量约20-24kDa）是信号转导的核心：

TCR信号枢纽：与TCRαβ/γδ链通过盐桥结合，胞内段含免疫受体酪氨酸活化基序（ITAM），TCR识别抗原后，ITAM磷酸化招募ZAP70，启动PLC-γ/NF-κB通路，是T细胞活化的“分子开关”；

免疫分型标志物：T细胞特异性表达（外周血T细胞95%阳性），联合CD4/CD8抗体可区分辅助性/细胞毒性T细胞亚群，在肿瘤微环境中CD3⁺T细胞密度与PD-1抑制剂疗效正相关；

治疗靶点价值：抗CD3-ε单抗可通过部分激活T细胞诱导免疫耐受，用于1型糖尿病、器官移植抗排斥治疗（NEJM, 2024）。

实验操作关键要点

流式细胞术（T细胞活化检测）：

样本处理：人PBMC经CD3/CD28磁珠激活72小时，CD3-ε抗体（APC标记）1:100稀释，4℃避光孵育30分钟，流式检测CD3⁺CD69⁺活化T细胞比例（较未激活组升高5-10倍）；

关键对照：同型对照（兔IgG-APC）确保非特异性结合率<2%，阳性对照选用Jurkat细胞（高表达CD3-ε）。

IHC-P肿瘤浸润T细胞计数：

样本：结直肠癌组织石蜡切片，CD3-ε在肿瘤间质淋巴细胞细胞膜呈棕黄色阳性，按“每mm²阳性细胞数”计算密度（>50个/mm²提示预后良好）；

抗原修复：EDTA缓冲液（pH9.0）高压修复15分钟，斯达特SDT-R137抗体1:2500稀释（5% BSA+TBST），4℃孵育过夜，DAB显色5分钟（控制背景）。

WB检测TCR复合物组装：

样本处理：Jurkat细胞用NP-40裂解液（含1mM Na₃VO₄） 冰上裂解30分钟，14000g离心20分钟取上清，抗体1:1000稀释，检测20kDa CD3-ε条带，同步验证CD3ζ链（16kDa）共表达。
