产品介绍This gene encodes a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus. This gene is expressed not only in T lymphocytes, but also in B cells, macrophages, and granulocytes. It is also expressed in specific regions of the brain. The protein functions to initiate or augment the early phase of T-cell activation, and may function as an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system. Multiple alternatively spliced transcript variants encoding different isoforms have been identified in this gene. [provided by RefSeq, Aug 2010].

靶点与功能

CD4（Cluster of Differentiation 4）是55kDa跨膜糖蛋白（免疫球蛋白超家族成员），核心功能与研究价值包括：

T细胞亚群标志物：特异性表达于辅助性T细胞（Th1/Th2/Th17）、调节性T细胞（Treg）及部分树突状细胞表面，流式检测中CD4⁺CD8⁻ T细胞占外周血T细胞的60-70%；

免疫共受体作用：通过胞外域D1区与MHC II类分子β2结构域结合，胞内段与Lck激酶关联，参与TCR信号放大，是T细胞活化的“分子开关”；

疾病关联：HIV通过CD4入侵T细胞导致免疫缺陷，而肿瘤微环境中CD4⁺ T细胞浸润密度与PD-1抑制剂响应率正相关（J Clin Oncol, 2024）。

实验操作关键要点

流式细胞术（T细胞亚群分析）：

样本处理：人外周血PBMC用PBS+2% FBS洗涤2次，CD4抗体（如Bethyl A700-015，PE标记）1:100稀释，4℃避光孵育30分钟，联合CD3-APC/CD8-FITC抗体，流式检测CD3⁺CD4⁺ T细胞比例（健康人参考范围30-40%）；

关键对照：同型对照（兔IgG-PE）确保非特异性结合率<2%，阳性对照选用Jurkat细胞（低表达CD4）和CD4⁺ T细胞系（如SupT1）。

IHC-P肿瘤浸润T细胞定位：

样本：非小细胞肺癌组织石蜡切片，CD4在肿瘤间质淋巴细胞细胞膜呈棕黄色阳性，计数阳性细胞密度（>5个/HPF提示预后较好）；

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟，3% H₂O₂室温封闭10分钟，抗体1:200稀释（5% BSA+TBST），4℃孵育过夜。

WB检测T细胞活化后表达变化：

样本处理：人CD4⁺ T细胞经抗CD3/CD28磁珠刺激72小时，RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心15分钟取上清；

结果判断：55kDa条带灰度值较未刺激组升高2倍（伴随CD69/CD25活化标志物上调），内参选用β-Actin（42kDa）。
