产品介绍The protein encoded by this gene is a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion and migration. It is a receptor for hyaluronic acid (HA) and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs). This protein participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, hematopoiesis, and tumor metastasis. Transcripts for this gene undergo complex alternative splicing that results in many functionally distinct isoforms, however, the full length nature of some of these variants has not been determined. Alternative splicing is the basis for the structural and functional diversity of this protein, and may be related to tumor metastasis. [provided by RefSeq, Jul 2008]

靶点与功能

CD44是细胞表面糖蛋白，分子量85-200kDa（因糖基化修饰差异），通过与透明质酸（HA）、胶原蛋白、金属蛋白酶等配体结合，参与：

生理过程：淋巴细胞归巢、细胞外基质黏附、胚胎发育；

病理意义：肿瘤侵袭转移（CD44⁺细胞被视为多种癌种的干细胞标志物）、炎症反应（如巨噬细胞募集）；

研究价值：区分肿瘤干细胞亚群（如CD44⁺/CD24⁻乳腺癌干细胞）、评估肿瘤微环境重塑能力。

实验操作关键要点

流式细胞术（FCM）检测肿瘤干细胞：

样本处理：单细胞悬液用含2% FBS的PBS重悬，抗体1:100稀释，4℃避光孵育30分钟；

对照设置：同型对照（小鼠IgG2b）、CD24抗体联合染色（区分CD44⁺/CD24⁻亚群）；

仪器参数：激发波长488nm，发射波长525nm（FITC标记）。

IHC-P组织染色（乳腺癌样本）：

抗原修复：柠檬酸钠缓冲液（pH6.0） 微波修复20分钟，冷却至室温；

阳性定位：肿瘤细胞细胞膜/胞质棕黄色着色，癌巢边缘侵袭区表达增强；

封闭处理：5% BSA+0.3% Triton X-100室温封闭1小时，减少非特异性结合。

WB验证（MCF-7细胞裂解液）：

样本制备：RIPA裂解液（含1mM PMSF）冰上裂解30分钟，12000g离心取上清；

电泳条件：8% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶；

抗体孵育：一抗1:800稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时。
