产品介绍The protein encoded by this gene is a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion and migration. It is a receptor for hyaluronic acid (HA) and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs). This protein participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, hematopoiesis, and tumor metastasis. Transcripts for this gene undergo complex alternative splicing that results in many functionally distinct isoforms, however, the full length nature of some of these variants has not been determined. Alternative splicing is the basis for the structural and functional diversity of this protein, and may be related to tumor metastasis. [provided by RefSeq, Jul 2008].

靶点与功能

CD44是一种透明质酸受体，通过选择性剪接产生标准型（CD44s）和变异型（CD44v）亚型，核心功能包括：

肿瘤进展：作为肿瘤干细胞（CSC）标志物，在乳腺癌、结直肠癌中高表达，与肿瘤侵袭转移及化疗耐药密切相关（临床样本显示CD44⁺细胞成瘤能力是CD44⁻细胞的5-10倍）；

细胞黏附：通过结合细胞外基质（ECM）中的透明质酸，调控细胞迁移和上皮-间质转化（EMT）；

免疫调节：参与淋巴细胞归巢及炎症反应，是T细胞活化和肿瘤免疫微环境重塑的关键分子。

实验操作关键要点

流式细胞术优化（肿瘤细胞分选）：

样本处理：单细胞悬液用含2% FBS的PBS洗涤2次，避免EDTA螯合剂影响抗原-抗体结合；

抗体孵育：推荐1:50稀释（每10⁶细胞加入10μL抗体），4℃避光孵育30分钟，无需洗涤直接上机（FITC标记可直接检测）；

阳性对照：选择MCF-7乳腺癌细胞系（CD44高表达），阴性对照使用同型IgG-FITC抗体。

IF双标实验（肿瘤干细胞定位）：

细胞固定：4%多聚甲醛室温固定15分钟，0.2% Triton X-100透化10分钟；

共染推荐：与CD24抗体（PE标记）联合使用，流式分选CD44⁺/CD24⁻表型的肿瘤干细胞亚群；

激光条件：FITC激发波长488nm，发射波长525nm；PE激发波长561nm，发射波长585nm。

IHC-P注意事项（临床组织样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强细胞膜信号；

阳性定位：肿瘤组织中细胞膜/胞质呈棕黄色阳性，正常组织中淋巴细胞和上皮细胞弱阳性；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性结合。
