产品介绍The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. Four alternatively spliced transcripts variants of this gene, which encode distinct isoforms, have been reported. [provided by RefSeq, Jul 2008].

靶点与功能

是跨膜蛋白酪氨酸磷酸酶，几乎在所有造血细胞（像白细胞、淋巴细胞等）表面都有表达，成熟红细胞和血小板除外。它的主要作用是通过去磷酸化调节免疫细胞的活化信号，就像一个“开关”控制着T细胞和B细胞的激活。而且它有不同的“变身”（剪接异构体），比如CD45RA、CD45RO，这些“变身”能帮我们区分不同状态的免疫细胞，比如初始T细胞和记忆T细胞，在免疫相关疾病研究和肿瘤免疫治疗中都特别有用。

实验应用

流式细胞术：用荧光标记的抗体，可以给免疫细胞“贴标签”，比如区分T细胞、B细胞，记得染色前用Fc受体封闭剂处理样本，能减少非特异性结合。

WB实验：检测细胞裂解液时，因为有不同异构体，WB结果可能会出现180kDa、190kDa、220kDa等多条带，这是正常现象。

IHC实验：在组织切片中，阳性信号主要在免疫细胞的细胞膜上，呈棕黄色，比如淋巴结组织中的淋巴细胞就会显色，而上皮细胞一般不显色，可作为阴性对照。
